产烟酸羟化酶菌株Pseudomonas sp BK-1培养条件的研究

Cultivation and biotransformation of Pseudomonas sp.BK-1

  • 摘要: 对产烟酸羟化酶的菌株Pseudomonas sp.BK-1的培养条件进行了优化,结果表明10g·L-1蔗糖为最佳碳源,20g·L-1玉米浆为最佳氮源,诱导物烟酸浓度12.5g·L-1,培养基的最适初始pH值7.0.5L发酵罐中进行发酵,在转速400r·min-1、通气量4.2L·min-1、30℃条件下,16h时酶活可达1.47U·mL-1.经36h的连续催化,产物6-羟基烟酸的浓度可以积累到154g·L-1.

     

    Abstract: To increase the activity of nicotinic acid hydroxylase of Pseudomonas sp.BK-1,the culture conditions of the strain were optimized.The experimental results show that 10 g·L-1 sucrose is the optimal carbon source and 20 g·L-1 corn steep liquor is the optimal nitrogen source.The optimized concentration of the inducer nicotinic acid is 12.5 g·L-1 and initial pH of culture medium is 7.0.Under the conditions of rotation 400 r/min and ventilate 4.2 L/min the highest enzyme activity of this strain can reach 1.47 U·mL-1 at 16 h in a 5 liter fermentor.The accumulation of 6-hydroxynicotinic acid in reaction mixture by Pseudomonas sp.BK-1 can reach 154 g·L-1 at 36 h.
